Abstract : Provides a basis and overall context for concepts introduced and discussed in more detail in subsequent chapters in this book. An historical context is given to the topic of drug discovery, followed by a high level overview of modern approaches to rational drug design. The approaches and steps typically encountered in modern rational drug design, including target identification, lead discovery, and lead modification, are introduced, and an overview of the subsequent steps required for drug development prior to regulatory approval, such as pharmacokinetics, preclinical studies, clinical trials, and intellectual property issues, are summarized.
